signaling pathways and DsbA-L.

OBJECTIVE: The natural polyphenol resveratrol (RSV) displays a wide spectrum of health

beneficial activities, yet the precise mechanisms remain to be fully elucidated. Here we show that

RSV promotes the multimerization and cellular levels of adiponectin in 3T3-L1 adipocytes.

METHODS AND RESULTS: The stimulatory effect of RSV was not affected by knocking out

Sirt1, but was diminished by suppressing the expression levels of DsbA-L, a recently identified

adiponectin-interactive protein that promotes adiponectin multimerization. Suppression of the Akt

signaling pathway resulted in an increase in the expression levels of DsbA-L and adiponectin. On

the other hand, knocking out FOXO1 or suppressing the activity or expression levels of the AMP-

activated protein kinase (AMPK) down-regulated DsbA-L and adiponectin. The stimulatory effect

of RSV on adiponectin and DsbA-L expression was completely diminished in FOXO1-suppressed

and AMPK-inactivated 3T3-L1 adipocytes.

CONCLUSIONS: Taken together, our results demonstrate that RSV promotes adiponectin

multimerization in 3T3-L1 adipocytes via a Sirt1-independent mechanism. In addition, we show

that the stimulatory effect of RSV is regulated by both the Akt/FOXO1 and the AMPK signaling

pathways. Last, we show that DsbA-L plays a critical role in the promoting effect of RSV on

adiponectin multimerization and cellular levels.

PURPOSE OF REVIEW: The present review is a critical examination of the most recent

published work on the role of zinc in the development and treatment of mood disorders.

RECENT FINDINGS: Clinical studies and experimental work using animal models have both

revealed a link between zinc status and neuropsychological disorders such as depression and

anxiety. Not only has zinc deficiency been shown to induce depression-like and anxiety-like

behaviors, supplementation has been used as a treatment for major depression. Zinc

administration improves the efficacy of antidepressant drugs in depressed patients and may have

a particular role to play in treatment-resistant patients. Recent investigations into the molecular

mechanisms responsible for these observations suggest a role for zinc in the regulation of

neurotransmitter systems, antioxidant mechanisms, neurotrophic factors, and neuronal precursor

cells.

SUMMARY: The data reviewed here not only indicate a role for zinc deficiency in the

development of mood disorders, but also show that zinc may also be important in their treatment.

Given the prevalence of zinc deficiency in human populations, this work has the potential to

influence strategies to prevent and treat these disorders.





XCAP’s Gian Villante: Football’s ‘10 Times’ More Dangerous Than MMA

After tryouts with various NFL teams didn’t work out, Gian Villante  decided to try something he sees as safer: MMA.

“I feel like football’s 10 times more dangerous and savage than MMA is,” Villante said Monday on the Sherdog Radio Network’s “Beatdown” show. “I’ve got 10 times worse injuries playing football than I did with MMA.”  Villante recently trained at Xreme Couture, MMA.

The 25-year-old native of Wantagh, N.Y., was a standout linebacker at Hofstra University. Since transitioning to fighting, he has compiled a 7-1 record and earned prospect status as Ring of Combat’s heavyweight champion. He fights Chad Griggs on Saturday in an alternate bout for Strikeforce’s heavyweight tournament.

“In [MMA], we have weight divisions. We see the shots that are coming,” Villante said. “In football, you don’t see it. It’s a 400-pound guy coming as fast as a car can move sometimes, and you don’t see him. He’s coming to clean your clock, and you’re not even looking at him. … That’s why these guys are getting these head injuries and shoulders are dislocating and all that stuff.”

Villante has cleaned a few clocks himself. All seven of his wins have been finishes, including a knockout via a head kick. Clearly the athleticism he used on the football field has translated to the ring.

“If we don’t feel like we’re getting the right opportunity somewhere, why not take your talent somewhere else? Football players, most of us are pretty good athletes,” Villante said. “Big, strong guys that are explosive, fast athletes. In mixed martial arts, that bodes well for knockout power.”

Of course, Villante was not just a football player. He also wrestled at Hofstra.

“I think wrestling’s very important too,” he said. “If you get a guy who just played football and has no wrestling experience, you’re going to have some trouble in there. I think that’s where it kind of helped me mix in and start off my career real fast and in the right direction.”

Villante has a two-year, six-fight deal with Strikeforce. Although he sees his future in the light heavyweight division, he said the alternate matchup against Griggs and a possible slot in the heavyweight tournament was too good to pass up.

“Chad Griggs isn’t a huge heavyweight,” Villante explained. “He’s 235, I believe. Nothing too big. He’s a guy that I think is a good fight for me.”

Griggs is coming off an upset win over Bobby Lashley last August. Villante was impressed with the fact that Griggs was not scared of his larger opponent.

“He’s a guy that comes to fight,” Villante said. “I’m not going to intimidate him if I flex at him because I guess Bobby Lashley is a little bigger than I am. … I think you can expect fireworks because we’re two guys that have pretty much finished every fight we’ve been in.”

Villante is expecting another finish and another win on Saturday as well.

“Like a football linebacker, if I see a hole, I’m going to go through. If I see an opening, if I see a guy hurt, I’m going to finish it,” Villante said. “If I get on him and I can catch him with something early, then I’m not going to take it easy. I’m going to try to finish the fight right away.”

XCAP Fighter Mike Pyle takes aim at John Hathaway at UFC 120

Xtreme Couture Athletic Pharmaceuticals (XCAP.tv)  fighter and UFC welterweight veteran, Mike Pyle has been speaking about his upcoming fight at UFC 120 this weekend with John Hathaway in London, England, with the man dubbed “Quicksand” saying he plans to live up to his reputation on the night.

Speaking via UFC.com, Pyle said:

“It doesn’t matter where it’s at – it could be here, it could be over there – I’m going in to do my job and that’s to go in there and beat him. That’s what I’m training for, and that’s what I’ve been eating, sleeping and breathing – going in there and competing at my best on that day against John Hathaway. So that’s what I’m gonna do. I feel good about it, I kinda like being the underdog, I like being the guy who’s gonna get booed and who nobody’s gonna like there, but I think I’ll capture some fans after it’s all said and done.”w

“That’s expected and that’s how this sport is gonna grow and I’m a part of this sport. Whether I like it or not, which I do, there are gonna be younger, stronger, faster guys coming in with more talent. That’s all there is to it, and why not pit yourself against them and just keep going until you just can’t anymore? It’s all good fun and I’m a fighter – whether you’re younger or older, I’m gonna fight you anyway and put myself to the test.”

“He’s a young guy coming in who’s eager, who’s trying to make a name for himself, and he is making a name for himself coming off a good performance against (Diego) Sanchez and he looked good doing it. But a lot of the fighters he’s faced have always been shorter and he’s never really had to deal with someone who was almost his length, so I look forward to the challenge. I think that I’m gonna have the edge on him. I’ve got him on experience and just sheer guts and toughness, and I think I’m an all-around more skillful guy than him when it comes down to it.”

“That’s the way that I like to fight and why I got the nickname “Quicksand” given to me. The harder you’re fighting in the quicksand, the worse it’s gonna get and you’re gonna keep sinking. I always try to set things up and I don’t try to bully my way into submissions because then they know it’s coming. And there’s no better way to get a nice, fast, and clean sub then to go with the flow and try to keep my technique sound where I know that if I pressure a guy into one certain way, he’s gonna go another way to get out, and that’s what I’m looking for. It’s like going fishing. I’m trying to outsmart him and hopefully I’ll hook him.”

In the opening match of the night Mike Pyle (19-7), a 3 time UFC veteran hopes to get back in the win column against the hard hitting Jesse Lennox (11-2). Both coming off losses, this bout is guaranteed to see a level of aggression from the two fighters that will all but guarantee an exciting contest.

“Camps been great, it went smooth. Nice and smooth and I am ready to fight. I have some bumps and bruises but I’m 100% healthy,” Mike Pyle disclosed to BJPENN.COM.

“Lenox is a tough kid; he puts up a good fight, so I’m looking forward to that challenge. I don’t really know where he poses the toughest challenge for me. We will just have to see, once the leather starts flying. We’ll just see where it goes from there. The main strategy is to go in and punch him in his face until he falls down and if that doesn’t work than I’ll move on to plan B. I’m just gonna go in there and give him hell, you can always have a game plan but as soon as you get hit, shit happens and things change, so I’m going to go with the flow and take the kid out.”

Pyle continued…

“In a perfect world I’d like to go in there and slip his big overhand right and take him down right into a choke, but that’s a perfect world. I’d like to just get in there and really showcase some skills that I haven’t got to in the past in the UFC and just really prove to myself that I belong there and that I’m ready to fight the best that’s in the UFC and that’s about it.”

Pyle is coming out of a great camp, has some of the best training partners in the business and an all-star cast working his corner. While, his Xtreme Couture coaches and teammates cannot go in there and fight the fight for him, Mike, believes that his team support is an advantage going into this bout.

“Kampmann, Tyson and I are a bunch of animals in there right now; we are so ready it’s not even funny. It helps out big time that they are both on the card with me. I got Ron Frazier, Randy Couture and Ray Sefo in my corner for this fight. Its awesome working with Randy, he gives you tons of confidence because he’s been there and done that.
